CAP EQA PNH results - problems with CD59

We have been subscribing to CAP PNH EQA for about 1 year.

Have encountered a problem in results for recent CAP PNH EQA that we are
not sure what to do.

In the recent March CAP PNH EQA, for PNH 01 sample, we could identify only
1 single population using IOTest CD59 (Clone P282E) although there is
supposed to be 2 populations..

The reason is  due to the antibody clone we use since  with  BD-CD59 or
Caltag-CD59 there is clear separation of  PNH cells ie. 2 populations seen
in contrast to	IOTest-CD59 which gives only 1 single population. We do  no
lyse, no wash staining, using IOTest CD59 (Clone P282E) .

Our normal approach for PNH screening  is CD16/55/59 on granulocytes and
CD55/59 on rbcs and we use IO reagents since we have use of  Coulter
machine on reagent contract basis. Seems to be working fine past few years
and we validate the reagents for each change of lot and ensure they are
used before expiry period.

In view of recent CAP results, we are wondering if we shoulcl change our
practice or  CD59 antibody clones we use
Just wonder what is anyone’s experience and response ??
