Hi Alessandro. We tested various fix&perm methods for staining of different intracellular antigens- also Ki-67- and compared different commercial available fix&perm kits with self-made fix&perm solutions. Our results obtained that for Ki-67 either PFA fixation in combination with saponin permeabilisation or the BD fix&perm kit gave the best results of the tested fix&perm methods- at least in the porcine system. Specified procedures and staining results of surface and intracellular antigens were published in Vet. Immunol Immunopathol. 2008 Feb 15;121(3-4):251-9. Good luck, Tobias Dipl.-Biol. Dear All Can anybody suggest which is the best method to permeabilize cells in order to obtain the most reliable staining with Ki-67 Ab from BD?
