Dear Colleagues, We have a user, who wants to measure the DNA of parasites (e.g C.elegans) using FACS Calibur or LSRII. However, when we run the sample we don't appear to detect any DNA profile, although the QC check samples (chicken erythrocyte nuclei and calf thymocyte nuclei from BD) show nice DNA and linear profiles. I would like to know if anyone has a protocol for preparing of nematode cells for FACS and can provide any suggestions for the setting of above instruments. Any guidelines would be helpful in this procedure. Thanks very much for your help.
