Hi Andy We have used fluorescent antibodies to differentiation antigens on haemopoeitic cells that are PKH26 labelled and all worked well. There was no need to modify the antibody reaction protocol. No idea about frozen tissues though. Hope this helps Best wishes Ian Ian Titley PhD Section of Haemato-oncology Institute of Cancer Research 15 Cotswold Road Belmont Sutton Surrey SM2 5NG UK Tel +44 (0)20 8722 4255 >>> Andy Hoffman <andrew.hoffman@tufts.edu> 06/06/2008 20:49 >>> Would anyone care to comment on incompatibilities that may arise between PKH staining and surface immunostaining for flow (other than usual conflicts in fluorochrome selection). For example does PKH have any affects on subsequent antibody binding and therefore modify antibody concentrations or incubation times/temp, etc? Related to this, if anyone has done concurrent PKH staining and immunostaining of frozen tissues (with or without a permeablization step) I would also welcome advice. Thanks so much!
