Hi Barb, I have a 130um nozzle and using a frequency between 17-18 at 12 psi works well for us. We can see the break-off and have no issues sorting. You may need to place the attenuation on. The key is to make sure you get the air out of system (let it sit 30 -60 mins before accudrop) and start with a full PBS container. Good luck, Greg ____________________________________________________________ Gregory Veltri Head of Flow Cytometry Cancer Research UK Cambridge Research Institute Robinson Way Cambridge CB2 0RE 01223 404170 http://science.cancerresearchuk.org/sci/flow_cytometry/ -----Original Message----- From: Barbara Joan Taylor [mailto:taylorba@mail.nih.gov] Sent: Thu 5/8/2008 8:44 PM To: cyto-inbox Subject: Aria and 130 nozzle Aria users I am going to try the 130 nozzle for the first time - can I get some suggestions for a starting point for sheath pressure and frequency?
