Dear Flow Experts: It is so good to know we are not alone! Thanks for all replies to my e-mail. As my last sorting was so long ago (more that 6 month I believe) I was so stressed! I run cell lines that were bigger than lymphocytes before using the 70um nozzle at medium pressure, so I have the same template with voltages for those cells, I change them a little but I didn't think it was that much change and that was my mistake!!! After I started sorting I realized that cells were going to the bottom so fast and shaking at 300 was not enough to keep them in suspension, plus all the "rainy side stream" and I didn't have too many cells to sort so to start again (changing nozzle, set the drop delay again and do the sorting with the new settings). I didn't know these cells were so big!!! I am sure we will not have any problem when I run them next time. I needed your support! This mailing list really works! Thanks a lot Dr Robinson for keeping it going. Thank you all: Charles, Steve, Pamela, Barbara, Dan, Jerry, Charles, Debora, Katherina, Joseph for your concern and prompt reply!!! All were coincident in their suggestions about the 100um nozzle and accudrop and also about running accudrop after preparation for asceptic sort and then run ethanol or bleach + sterile PBS to ensure sterility.
