Calcium measurement - dye loading question

Hi all,


I have an investigator wanting to do some calcium measurements on
macrophages.  I have scanned the literature and there seems to be several
additives to help load the dyes or prevent them from being pumped out.	I
have found Probenicid, Pluronic F-127, PowerLoad and Sulfinpyrazone alone or
in combination mentioned.  Does anyone have any experience which works best,
so we could maybe save a little time and money trying to figure it out?
Also, does it matter what form you buy - water soluble or in DMSO?  We will
be trying the ratio metric measurement of fluo-3 and Fura Red AM on a
FACSAria.  Any other suggestions are welcome.
