Hello Flowers, I have a student interested in sorting marine bacteria isolated from coral samples. We performed a sort using PBS as the sheath fluid and found that we had low recovery of RNA and DNA. We are suspecting that the bacteria are being affected by the change in osmolarity between the seawater and the sheath fluid. We have a couple of ideas as to how to solve this: We could change the sheath fluid to 3.5x PBS, approaching osmolarity of seawater. So the question is, has anyone had experience doing this kind of sort? Do you think that changing the PBS concentration would affect sort conditions? Any suggestions would be greatly appreciated.
