analysis of mitochondrial size and density with Aria

Dear all:

We are analyzing mitochondrial size and density using a FACSAria.
However, the distribution patterns on FSC/SSC from the Aria (see the
attached) are different from what were published in the literature using
either the FACScan or FACSCalibur (e.g., Mattiasson, Cytometry Part A
60A:145–154, 2004, showing SSC in logarithmic scale and Lecoeur et al,
Exp Cell Res 294:106-17, 2004, showing SSC in linear scale). In
addition, we are not quite sure which ND filter we should use for our
analysis. It appears that ND filter selection also affects the FSC/SSC
distribution patterns. 

We stain our mitochondrial preparations with NAO or JC-1 to confirm that
they are true mitochondria (>90% positive). We also confirm the quality
of the preparations by measuring their oxygen consumption and routinely
have RCR >4. Thus, these are true mitochondria with fine quality. Yet…
