cell counts post sort, low cell numbers

Hello all,

I would like to add to my previous synopsis of cell
counting methods appropriate for verifying cell counts
after sorting very few cells. The investigator here
found a method that is working pretty well for him:

 He found some Teflon Printed Slides (24-well, 4mm
well diameter) cat# 63430-04 from Electron Microscopy
Sciences. The teflon circles hold small amounts of
liquid in place. He adds 1ul of trypan blue and 1ul of
the cell suspension from the sort tube. He counts all
cells present, and this is the concentration of
cells/ul. From this he can calculate total number of
cells. For more accuracy, you can do more than 1 well
and average. Depending on the sort population he
usually gets counts of about 70-90% of the counts
reported by the ARIA. He feels these counts give him a
better estimation of how many viable cells he is
working with in the tube, so he can plan his
injections with more accuracy.
