Co-stimulation for intracellular cytokine detection

Dear All,

I would like to know if anyone has any experience of the effects of 
co-stimulation using anti-CD28+anti-CD49d (or other combinations) on 
intracellular cytokine staining. We are characterizing	human PBMC 
immune responses to a crude microbe-derived antigen (containing 
proteins, not peptides). We have observed that the detection of 
cytokines (like IFN-gamma) is not much affected by the presence or 
absence of the added -stimulatory antibodies. Therefore, we are 
interested to know if any Flower has experience of such 
co-stimulation in similar systems and could summarize the pros and 
cons of this type of co-stimulation.
