The first reported dual-wavelength immunofluorescence flow cytometry experiment - in which compensation was first described (Loken MR, Parks DR, Herzenberg LA: Two-color immunofluorescence using a fluorescence-activated cell sorter. J Histochem Cytochem 25:899, 1977) - used fluorescein and rhodamine as labels and excited with the 515 nm green argon line. This was suboptimal for both dyes, but there is virtually no excitation of rhodamine at 488 nm. As long as you use appropriate filters, you shouldn't have a problem with a green laser. These days, a 532 nm YAG is probably the most practical laser to use, and you probably won't need more than 50-100 mW. -HowardReceived on Mon Dec 5 12:38:00 2005
