Dear Ramunas, All Treg cells have to have CD4 and CD25. CD25 can be expressed AFTER stimulation of CD4 cells, and it is only the cells which are CD4+CD25+ at rest, which are called Treg cells. The only problem is that some T cells are in a state of 'stimulation' even at rest, so it is possible to still overestimate Treg cells by measuring CD4+CD25+ cells. Hope this helps.
