Like most Abs, tetramers have to be titrated when staining PBMC. Recommendations from commercial vendors indicate that one should use "X" dilution per 10^6 cells. Does that mean 10^6 positive cells (in the unlikely event that all cells would be tetramer positive), or does it mean 10^6 total PBMC with a small percentage of PBMC likely to be tetramer-positive? Tetramers are so expensive that I don't want to waste the reagent if I'm going to stain 40 x 10^6 PBMC when only 1.5% of them will be positive.
