Dear flowers I wondered if you can help.... I want to purchase three (ideally four) highly cross adsorbed secondary antibodies recognising goat, rabbit and mouse with conjugates such that I can correlate data that I obtain on a FACS Calibur with a Zeiss axioplan microscope (mercury short arc lamp). I would like at least one of the combinations (mouse and the rabbit preferably) conjugates to FRET. Our (current) filter sets on the FACS are FL1; 530/30 BP FL2; 585/42 BP FL3; 670LP FL4; 661/16BP Our Zeiss microscope filter sets are 1. Zeiss Filter set 10 : Ex 450-490: dichroic 510 Em: 515-565 2. Zeiss Filter set 37 : Ex 450/50BP: 480FT: Em: 510/50 3. Zeiss Filter set 15 : Ex 546/12BP: 580FT: Em: 590LP I could buy another one/(two) filter sets (to give us four colours) to give us channels on the microscope equating to FL3 and FL4. I'm trying to figure this out myself but I could really use some expertise.... Can any of you clever people suggest some appropriate conjugate combinations or even antibodies?
