We have found that Pacific Blue is brighter than either Cascade Blue or Alexa Fluor 405 dye. Also, in some preliminary work, Cascade Yellow gives very reasonable resolution when read at 585 nm rather than at 530 nm, with very little spectral overlap from Pacific Blue. We are wanting to get a dye that is excited at 407nm and is as bright as possible (when conjugated to an antibody with MP's Zenon reagents). The current PMT filters are at 530/30 nm and 450/40 nm. We could change those if that would help. We have tried Alexa 430 and found it to be rather dull. Any thoughts about whether Alexa 405 or Cascade Blue would be any brighter? Thanks. Alice Alice L.
