Is fixation needed for IC staining? Detecting F-cells

Hello fellow flowers-
I am looking to detect F-cells via flow.  I have read many papers indicating that you can
do this by IC staining with anti-HbF.  I have only read and performed IC staining
following some sort of fixation (i.e. gluteraldehyde or formaldehyde) and
permeabilization.  I have 2 questions:
1. Is is necessary (and if so, why) to fix cells before permeabilization?  ( I believe
the answer to to perserve cell morphology and prevent "leakage" of IC proteins, but I
wonder if there is another more important reason).
2. Has anyone heard of another method of detecting F-cells beyond anti-HbF IC staining?
Any cell surface marker known specifically for F-cells?
I am interested in performing cell sorting and isolating proteins,thus the fixation
process will be a huge issue in down stream applications.
