Fresh apoptotic PBMC are easily detectable by TUNEL assay(e.g. Gorczyca
et al.,Induction  of DNA strand breaks associated with apoptosis during
treatment of leukemias. Leukemia, (Baltimore) 7: 659-670, 1993).
Likewise, the activation-induced apoptosis of lymphocytes is best
detected using the TUNEL assay (e.g. Halicka et al., Enhancement of
activation-induced apoptosis of lymphocytes by the cytotoxic
ribonuclease onconase (Ranpirnase). Int. J. Oncol., 21:1245-1250, 2002).
However since no apoptotic cells are present among PBMC just after their
isolation from normal healthy individuals, therefore no TUNEL positive
cells can be detected unless one induces apoptosis e.g. by culturing for
3-5 days

Have you ever examined apoptosis in fresh PBMCs? I read in some 
articles that apoptosis in fresh PBMCs can not be detected by Tunel 
test; the cells should be cultured for 3-5 days before apoptosis test.
What is your idea? I would be grateful if you could kindly give me the 
benefits of your experiences.
