Hi Liza, Have a look at this: J Immunol. 1995 May 1;154(9):4294-301. Flow cytometric analysis for cytokine production identifies T helper 1, T helper 2, and T helper 0 cells within the human CD4+CD27- lymphocyte subpopulation. Elson LH, Nutman TB, Metcalfe DD, Prussin C. They describe a protocol for cryopreservation. good luck Mara ******************************* Dr. Mara Rocchi Ph.D. Flow Cytometer Manager Moredun Research Institute Pentland Science Park Bush Loan, Penicuik EH26 0PZ Scotland, UK Tel: +44-131-4455111 Fax: +44-131-4456111 e-mail: mara.rocchi@mri.sari.ac.uk -----Original Message----- From: lmcguire [mailto:liza.mcguire@mail.tju.edu] Sent: 23 December 2003 02:09 To: cyto-inbox Subject: Intrarcellular staining on thawed lymphocytes Hi flowers, Does anyone have experience with evaluating intracellular staining on thawed human lymphocytes? We currently stain fresh cells with Perforin but would like to be able to save the specimens received for future evaluation by cryopreserving them and later on thawing them and staining with Perforin or other cytokines. Is this achievable? What kind of problems can we expect? Has anyone done any recovery analysis, post thaw, to identify any obvious loss of cell surface epitopes? We will be staining with a couple of cell surface markers in addition to the intracellular staining. I appreciate any and all suggestions, information, guidance. Thank you for your help. <http://www.smileycentral.com/?partner=ZSzeb001> Happy Holidays <http://www.smileycentral.com/?partner=ZSzeb001> Liza McGuire Cancer Center Regulatory & Compliance Officer Thomas Jefferson University Hospital 125 South 9th Street 801 Sheridan Philadelphia, PA 19107 Phone: (215) 955-9384 Fax: (215) 955-9641 **************************************************************************** **************************************************************************** *********** The contents of this e-mail message and any attachments are private and confidential communications intended solely for the addressee(s) named in this message.
