Michael - we use TOPRO-3 as a vital dye at final concentrations between 1 and 5 nanomolar. The 1 millimolar (in DMSO) stock that comes from Molecular Probes lasts over a year at -20. For use, we make up a 5 micromolar solution in PBS (not FACS buffer). We've noticed that this solution lasts for 1-2 months at 4C, so we only make 1 mL or so at a time. This is now your 1000-5000X stock. You can dilute this further in PBS immediately before use, such that you're adding 10-50 microL to ~0.5 mL cell suspension (which can be in FACS buffer).
