Dear Flow-ers,

My colleagues encountered a suspected leukaemia case.

The results are as follows:
Female, aged 45, Bone marrow specimen with many blasts. About 60% of the
gated blast population (side scatter vs CD45) were:
CD34(96%), CD33(25%), DR(99%),CD13(6%), CD33(25%),CD117(3%),CD14(2%)
CD10(97%), CD19(99%), CD20(18%),CD22(72%), CD56(2%), CD2(1%), CD3(3%),CD7(9%)
For cytoplasmic staining results (by Fix and Perm):
MPO(26%), Tdt(45%), CD22(2%).
As a whole, the results is suggestive to be a common ALL case. However, the results of surface and
cytoplasmic CD22 are confusing to me.

Question: Why the cytoplasmic staining shown a obvious decreased of CD22 expression?
In my concept, it is the 'total' CD22 expression after intraprep. The real cytoplasmic expression is the 'total' minus 'surface' expression.
How come the surface CD22 (72%) is greater than 'total' CD22 (2%)? Is it the deterioration of CD22 antigen after 'Fix and Perm'
or simply technical error? Please feel free to give me any explanation and solution. Thanks.
